An Immunohistochemical Study on localization of Growth Hormone, Prolactin and ACTH in Pituitary Adenoma


Abstract
A new era began to merge in endocrinology with accurate assessment of the functional capacity of the pituitary gland by radioimmunoassay. Before the introduction of immunoperoxidase method, histopathological evaluation did not give to aid in solving pituitary related diagnostic problems. The author had been performed an experiment using immunoperoxidase method on surgical specimens of pituitary adenomas to compare the results from the serum assay of hormones.
Antihuman antibodies of growth hormone(GH), prolactin(PRL), and adrenocorticotrophic
hormone(ACTH) used were made from Dako Ltd. The results obtained were as follows;
1. By radioimmunoassay, 18 cases of adenoma were consisted of PRL producing adenomas 8 case, GH producing adenomas 4 cases, ACTH producing adenoma 1 case, GH and PRL producing adenoma 4 case, and adenoma unassociated with endocrine activity 1 case.
2. By immunoperoxidase method, 18 cases adenomas were consisted of GH producing adenomas; 3 cases, PRL producing adenomas; 3 cases, ACTH producing adenoma; 1 case, GH & PRL producing adenomas 6 cases, PRL & ACTH producing adenoma 1 case, GH, PRL, & ACTH producing adenoma 1 case, and adenoma without reaction to antihuman antibodies of GH, PRL, and ACTH: 3 cases.
3. In comparison between the results by radioimmunoassay and immunoperoxidase, only 7
of 18 cases were coincided and 11 of 18 cases revealed lack of an agreement.
With the above results, it is recommanded to report the pituitary adenoma as"¡¥producing adenoma with hyper¡¥emia," because the both results by radioimmunoassay or immunoperoxidase method means not always equal sense.
